Bottom line

One of the more credible dietary longevity candidates: it's already in food, animal data are strong, and early human trials on autophagy and memory are encouraging — but the large confirmatory RCTs aren't in yet.

What the evidence says

Spermidine stands out among 'natural' longevity compounds because the case is unusually coherent: it extends lifespan in multiple animal models, higher dietary intake tracks with lower mortality in population studies, and small human trials point to effects on autophagy markers and cognitive performance in older adults at risk of decline. It's graded B rather than A because those human trials are still small and short, and hard endpoints remain unproven — but the direction of evidence is more consistent than for most supplements.

Mechanism

Induces autophagy, in part by inhibiting acetyltransferase activity and promoting deacetylation of autophagy-related proteins. Also supports mitophagy and proteostasis and may improve mitochondrial and cardiovascular function — mechanisms that overlap with caloric restriction.

Safety

A normal component of the diet (wheat germ, legumes, aged cheese, mushrooms), so background exposure is universal and tolerability at dietary and low-supplement doses is good. Long-term high-dose supplementation is less characterised, but no major safety signal has emerged at studied doses.

Dosage context

Human studies typically use ~1–6 mg/day, often as a standardised wheat-germ extract. This overlaps with what a spermidine-rich diet already provides, which is part of the appeal — but also means 'more is better' hasn't been demonstrated.

Among the polyamines, spermidine has the most promising human base — which is exactly why we resist overselling it. B means 'worth watching and reasonable to get from diet', not 'proven pill'.
